Life is dependent on the preservation and storage of information. The genome and epigenome are the two central storehouses of information in eukaryotes, and although they work interdependently, they are fundamentally quite different. Genetic information is consistent across all body cells throughout the life of an individual while epigenetic information varies between cells as well as changes over time and as per environment.
Researchers have identified several hallmarks of aging such as epigenetic alterations, genomic instability, cellular senescence, telomere attrition, mitochondrial dysfunction, and others [1]. These are known to play a role in the dysfunction and deterioration of cells with age. David Sinclair and other researchers have previously indicated that loss of epigenetic information can cause changes in gene expression, leading to cellular identity loss. Previous studies in mice have also shown that cell injuries such as cell crushing and DNA double-strand breaks can promote loss of epigenetic information which can accelerate aging along with age-related diseases [2].
Cellular senescence is a state of stable cell cycle arrest that can be triggered due to a wide range of extrinsic as well as intrinsic factors. It promotes tissue remodeling, wound repair, and cancer prevention by stopping the proliferation of damaged and aged cells. Senescent cells are characterized by metabolic and morphological alterations, reorganization of the chromatin, and release of pro-inflammatory substances known as the senescence-associated secretory phenotype (SASP) [3]. Irreparable DNA damage, loss of epigenetic information, and telomere shortening are a few factors that can initiate cellular senescence. Accumulation of senescent cells with age results in inflammation as well as the generation of reactive oxygen species (ROS).

Hello, i am back! Blood, sweat and tears have been shed these past months of absence. But enough sweating was done during the summer, tears have already been shed, so that just leaves me with blood. And whether taking a part of blood (plasma) and injecting it into old animals is enough for rejuvenation. That’s right, many of you may remember, earlier this year i made a video on the oldest living rat.- and now we have some updates: in particular, (i) what actually were the rats given, and (ii) what changes were seen, showing both some DNAm and glycan age data.

Let’s say that it is a curse. The issue is he is also against life extension entirely. Maybe I want 200 years. Or 1,000. I have zero concern over a boredom problem as it is brain process which can eventually be controlled. And I am disgusted with the idea that I have to die because we might not progress very fast? Ugh.
Elon Musk has said a lot of potentially stupid stuff about aging and longevity, from saying that people shouldn’t live very long because society would ossify to advocating that we judge people based on their chronological age. Most recently, he’s taken to Twitter (aka X) to say “May you live forever is the worst possible curse once you understand deep time.” In this case though, he’s not wrong.

For the past five years, Silicon Valley biotech Rejuvenation Technologies has been quietly working on a therapeutic platform to extend telomeres in the human body, with the goal of boosting longevity and healthspan. Yesterday, the company emerged from stealth with a healthy seed funding round of $10.6 million, led by Khosla Ventures.
Rejuvenation has developed a synthetic mRNA-based approach to restoring telomeres to a “healthy length” – capable of reversing a decade of telomere shortening in a single dose. The mRNA produces telomerase, an enzyme that plays a critical role in maintaining the length of telomeres. Following positive preclinical results in lung and liver disease indications, the company is now preparing the path towards its first in-human trials.
Longevity. Technology: Telomeres are protective structures on our DNA that prevent the loss of genetic information as the cells in our bodies divide. With each cellular division, our telomeres gradually shorten and, when they become critically short, our cells enter a state of senescence or die. Research has shown that telomere length is closely associated with lifespan and healthspan, and telomere shortening is recognized as one of the primary hallmarks of aging.
While it has long been known that the enzyme telomerase extends telomeres, it was only recently that safe, rapid telomere extension has been enabled by the delivery of mRNA. To learn more about how Rejuvenation Technologies is targeting telomeres and longevity, we caught up with co-founders Dr Glenn Markov and Dr John Ramunas.
